Snippet: After lysis of red blood cells, peripheral blood mononuclear cells (PBMCs) were stained with fluorophore-conjugated antibodies specific for CD4, CD5, CD8, CD79b and FoxP3 and analyzed by flow cytometry to identify major lymphocyte subsets. Document: After lysis of red blood cells, peripheral blood mononuclear cells (PBMCs) were stained with fluorophore-conjugated antibodies specific for CD4, CD5, CD8, CD79b and FoxP3 and analyzed by flow cytometry to identify major lymphocyte subsets. Total RNA was extracted from PBMCs for quantitative reverse transcriptase polymerase chain reactions using primers for three reference genes and for interferon gamma (IFNc) and interleukin (IL)-10. The serum concentrations of five cytokines (IL-2, IL-6, IL-8, IL-10 and TNFa) were measured using enhanced chemiluminescent assays. Results of complete blood cell counts and serum biochemical profiles were recorded for all INF and IMHA cases.
