The pcDNA3.1-M-rvfv plasmid was digested with the Not I and Kpn I enzymes and evaluated by performing agarose gel electrophoresis (Fig. 2) . The results showed two nucleic acid bands, at approximately 3200 bp and 5500 bp, which were consistent with the target gene fragment (3218 bp) and the pcDNA3.1 empty plasmid (5428 bp), respectively. This result verified the correct insertion of the target fragment. The gene fragment obtained in this experiment and the target gene sequence obtained by using gene sequencing were 100% homologous, indicating that the pcDNA3.1-M-rvfv recombinant plasmid was correctly constructed.
