miRNomes of haematopoietic stem cells and dendritic cells identify miR-30b as a regulator of Notch1

Next, we examined the roles of Notch1 in cytokine production in DCreg, and we found that inhibition of Notch1, just like miR-30b overexpression, increased IL-10 and NO production in DCreg, and Notch1 ligand, which activates Notch1 signaling, repressed IL-10 and NO production in DCreg (Fig. 6g) . These results further confirm the enhancing effect of Notch1 signaling on IL-10 and NO production in DCreg. Similar results of IL-10 production were observed, as determined by intracellular cytokine staining (Fig. 6h) . Additionally, we found that the enhanced Notch1 expression was significantly downregulated by Notch1-specific small interfering RNA (siRNA) once miR-30b was inhibited. (Fig. 7a) . Notch1-specific siRNA could rescue miR-30b inhibition-induced decrease of IL-10 and NO production in DCreg (Fig. 7b) . Similar results of IL-10 production were also observed in the experiments with intracellular cytokine staining (Fig. 7c ).
