Document: Results of this preliminary study document significant, repeatable hemodynamic alterations associated with intravenous CoCl 2 administration to horses. Further, the degree of hypertension observed following infusion raises humane and human safety concerns if doses of >1 mg/kg are used. The study objective was to evaluate the effects of patient age on aminoglycoside pharmacokinetics in young-adult (5-10 years) versus geriatric (≥25 years) healthy horses, receiving a single 6.6 mg/ kg intravenous injection of gentamicin (Henry ScheinÒ). Heparinized plasma samples were obtained at designated time-points following drug administration and frozen at À80°C until assayed by a validated immunoassay (QMSÒ). Gentamicin plasma concentrations versus time plots were analyzed using a two-compartment model and commercial software (WinNonlin-v6.4). Data were compared between age-groups using independent samples t-test; P < 0.05 considered significant.
