The sense primer had an EcoRI site engineered into its 5' end and the antisense primer a Sail site at its 5' end. The product was purified from the reaction mixture by phenol:chloroform extraction, digested with EcoRI and Sail, purified again and ligated into the EcoRI/SalI site of pSVK3.
