Title: Chronic hM3Dq signaling in microglia ameliorates neuroinflammation in male mice Document date: 2020_1_28
ID: ely200x3_8
Snippet: Using Cre/lox technology (Parkhurst et al., 2013) we generated a mouse line in which hM3Dq (Zhu et al., 2016) is selectively expressed in microglia. We found that acute activation of the hM3Dq pathway in microglia increased phagocytosis, increased the production of pro-inflammatory cytokines in the brain, but did not cause overt behavioral changes. Document: Using Cre/lox technology (Parkhurst et al., 2013) we generated a mouse line in which hM3Dq (Zhu et al., 2016) is selectively expressed in microglia. We found that acute activation of the hM3Dq pathway in microglia increased phagocytosis, increased the production of pro-inflammatory cytokines in the brain, but did not cause overt behavioral changes. In contrast, chronic activation of this signaling pathway attenuated the increase in brain cytokine production and sickness behavior triggered by low dose of lipopolysaccharide (LPS) injected peripherally. Our experiments provide a proof of principle that chronic M3 muscarinic Gq-linked signaling, and possibly other Gq-coupled GPCRs, can trigger immunological memory that helps to decrease inflammatory responses in the brain.
