Title: A Role for IFITM Proteins in Restriction of Mycobacterium tuberculosis Infection Document date: 2015_10_22
ID: zy82bo7q_35
Snippet: Lentiviruses were generated by transfecting 293T cells with the plasmids above in combination with the packaging plasmid psPAX2 and the envelope plasmid pMD2.G using Effectene transfection reagent (QIAGEN). Supernatants were collected 24 and 48 hr posttransfection, clarified by centrifugation, and stored at −80°C. Supernatants were collected 24 and 48 hr posttransfection, clarified by centrifugation, and stored at −80°C. THP-1 cells or A549 cells were transduced with lentiviral particles in the presence of 8 μg/ml polybrene (Millipore) and spinoculation. Transduced cells were selected and expanded by treatment with 1 μg/ml puromycin or 10 μg/ml blasticidin. THP-1 cells stably transduced with lentiviruses encoding shRNAs against MyD88, IRAK1, or TRAF6 were described previously (Ranjbar et al., 2012) .
