Snippet: Target identification is performed by evaluating the capacity of various C-depleted sera to restore the hemolytic capacity of the CHCP-treated serum. The optimal concentration of CHCP was 1.4 mg/mL, being just sufficient to completely inhibit the hemolytic activity of 1:8 diluted NHS through CP. 200 μL sensitized erythrocytes and 200 μL individual C-depleted serum were added to 200 μL sample-treated NHS, and the mixture was incubated at 37°C .....
Document: Target identification is performed by evaluating the capacity of various C-depleted sera to restore the hemolytic capacity of the CHCP-treated serum. The optimal concentration of CHCP was 1.4 mg/mL, being just sufficient to completely inhibit the hemolytic activity of 1:8 diluted NHS through CP. 200 μL sensitized erythrocytes and 200 μL individual C-depleted serum were added to 200 μL sample-treated NHS, and the mixture was incubated at 37°C for 30 min. After centrifugation, the percentage of hemolysis was calculated. The capacity of C-depleted sera to lyse erythrocytes was also assessed by incubating C-depleted serum directly with erythrocytes under the same conditions. All controls (vehicle, 100% lysis, complement and sample control) were treated in the same way.
