Cells were then pulsed with [ 3 H]-Leu (104 Bq/mL) for 12 h before the end of the treatment. At the end of the incubation, the cells were washed three times with ice-cold PBS and then disrupted by the addition of 200 µL 0.1% sodium dodecyl sulfate and 0.1 N NaOH. The solubilized cell lysates (100 µL) were added to 5 mL scintillation fluid. The incorporation of [ 3 H]-Leu into the protein was determined by scintillation counting. Incorporation is reported as the ratio: [ 3 H]-Leu incorporation/number of cells.
