All subsequent Tac-TGN38 chimeric and mu-tant DNAs were constructed by PCR, according to the procedure of Higuchi et al. (1988) and also cloned into pCDMS. The sequence of all the recombinant constructs was confirmed by the dideoxy chain-termination method.
