Since we wished to only use mutations that were synonymous in ORF2, we were unable to mutate the ORFX AUG codon in any other way. When utilized as an initiation codon, ACG is expected to be decoded as methionine by initiator Met-tRNA. Therefore the AUGm mutant is still expected to produce wt XP, albeit at a greatly reduced level.
