Among them, MDM2 is the major ubiquitin E3 ligase that leads to degradation of p53 by proteasome. Interestingly, the expression of MDM2 is induced by p53, thus forming a negative feedback loop for down-regulation of p53 (Ashcroft and Vousden, 1999; Oliner et al., 1992; Wu et al., 1993) . Under stressed conditions, however, the interaction of p53 with MDM2 and other negative regulators is disrupted by phosphorylation and acetylation, leading to stabilization and activation of p53.
