In the first patient, this mutation was found in a throat swab while a sputum sample from the same day still showed the original allele, 6446G. The SNP was analyzed by RT-PCR and Sanger sequencing in all sequential samples available from that patient ( Table 1) . The presence of separate genotypes in throat swabs and sputum strongly supported our suspicion of independent virus replication in the throat, rather than passive shedding there from the lung.
