Cell lines were cultured as previously described. Cell viability was assessed using a colorimetric assay (alamarBlueÒ). Cells were seeded in 96-well plates at a density of 2 x104 cells per well in cell culture medium containing a concentration range (0, 0.1, 1 and 10 µM) of the PKI tested and incubated for 48 h at 37°C, 5% CO2. AlamarBlueÒ (resazurin) was added 3 h prior to analysis and the reduced fluorescent molecule (resorufin) was measured by light absorbance in a fluorescence spectrophotometer (560EX nm/590EM nm). Experiments were performed in triplicate.
