Document: Trans‐4‐hydroxyl‐1‐proline, a nonproteinogenic amino acid, is an important constituent of the structural protein collagen. Increased trans‐4‐hydroxyl‐1‐proline concentrations in urine or serum are associated with degradation of connective tissue. Recently trans‐4‐hydroxyl‐1‐proline has been used as a biomarker of hepatic fibrosis in humans. The objectives of this study were to develop a liquid chromatography tandem mass spectrometry (LC‐MS/MS) method for the quantitative determination of endogenous trans‐4‐hydroxy‐l‐proline in dog serum, to analytically validate this method, and to asses serum trans‐4‐hydroxyl‐1‐proline concentrations in dogs with chronic hepatitis.
