A chimeric protein including a Kre2p cytoplasmic tail and enzymatic domain (KD-K) was properly targeted. In contrast, a luminal reporter protein could only be properly retained in the Golgi complex by the Kre2p cytoplasmic tail, plus the membrane-spanning and a 36amino acid residue stem domain (KKKP). The entire Kre2p cytoplasmic tail and membrane-spanning domain were not sufficient to retain Pho8p in the Golgi complex (KKP). Therefore, it appears that a combination of Kre2p topological domains is needed to achieve proper Golgi localization.
