Snippet: The cDNA encoding the human ␣2,3 sialyltransferase tagged with a 30-bp stretch of DNA encoding the 9E10 epitope of the myc protein was cloned into a mammalian expression vector (pcDNAI neo). At the COOH terminus of this type II transmembrane protein, the 9E10 epitope is far away from the membrane-spanning domain that has been shown to be involved in the localization of glycosyltransferases to their correct compartment within the Golgi (Machamer.....
Document: The cDNA encoding the human ␣2,3 sialyltransferase tagged with a 30-bp stretch of DNA encoding the 9E10 epitope of the myc protein was cloned into a mammalian expression vector (pcDNAI neo). At the COOH terminus of this type II transmembrane protein, the 9E10 epitope is far away from the membrane-spanning domain that has been shown to be involved in the localization of glycosyltransferases to their correct compartment within the Golgi (Machamer, 1993) . The construct was transfected into the breast carcinoma cell line, T47D, and two stable cell lines (designated T47D 3ST 2 and 3) were selected in the presence of gentimycin. The MUC1 mucin produced by the parental T47D cells carries short O-glycans that do not contain core-2 structures (Lloyd et al., 1996) .
