Previously, this amplification method was used for cellular transcriptome analysis (26, 27) with input amounts of 500 pg to 100 ng of total RNA. Here, using only femtograms to attograms of viral RNA with this amplification method in combination with Illumina sequencing and de novo assembly of viral reads, we successfully generated consensus sequence for the complete, or very nearly complete, CDS of viral genomes.
