The starting value for z is set to the median of the first ten data points in the time course. Since the measurement started before transfection, the first data points do not contain protein fluorescence but only background. The starting values for the expression rate and the mRNA degradation rate were set to 0 TL =1000 and =0.02, respectively. The maturation rate and the protein degradation rate were kept fixed to =1.206707417 and =0.004110525, respectively, during fitting.
