The scanning time-lapse measurement sequentially yields fluorescence image stacks from typically 70-100 positions. The movies are further processed to retrieve single-cell protein expression time courses as shown in Figure 1B . Each single-cell time course consists of the mean eGFP fluorescence intensity over the square occupied by a cell during the measurement period resulting in the protein expression kinetic for the respective cell.
