This second standard curve is used to determine the number of proviruses per reaction in preamplified dilutions of unknowns. To the extent that they are present, nonintegrated HIV sequences in the unknowns are part of the bulk HIV copy number and can be amplified linearly by gag priming in the preamplification. To correct for these, the signal contributed from unintegrated DNA is rou-tinely subtracted from the total signal by including a gag-only linear preamplification control (as elaborated below in Fig. 5) .
