After 24 hours, the co-culture from CD8+ T cells were stained with anti-CD69 antibody (BD #562884) and analyzed by flow cytometry (BD LSR II), and also imaged through the spinning disk confocal microscope (Nikon Yokogawa CSU-22). After 48 hours, cytokine concentration in the supernatant was measured by IL-2 ELISA (for CD4+ T cells, eBiosciences #BMS2221HS) and Interferon-γ (IFN-γ) ELISA (for CD8+ T cells, ThermoFisher #KHC4021).
