This is because pooling, if uncompensated, will dilute the presence of RNA/cDNA by a factor of b, which is expected to delay threshold-crossing. For example, if viral RNA is diluted by a factor of 8 from the original concentration in a sample then it will take 3 cycles of doubling in order to reach the original concentration. This should be taken into consideration when selecting maximal batch size.
