Three of the lungs were normal and three of the lungs met ARDS criteria before they were harvested. After cold preservation at 4°C, the right middle lobe was selected for cell isolation if no apparent signs of consolidation or hemorrhage by gross inspection were seen. The cell suspension was further incubated on Petri dishes coated with human IgG antibodies (Sigma) overnight at 37°C to remove the remaining macrophages and fibroblasts. Cell viability was assessed by the trypan blue exclusion method. AT2 cell purity was evaluated by Papanicolaou (Pap) staining.
