The experiments also included a non-template control and a positive control for the E-gene amplification. These samples were subject to the treatment that provided the results with the smallest cycle threshold (Ct) deviations from the standard protocol in the exploratory stage. The experiment was blind to the COVID-19 diagnosis assignment. Then, the resulting confusion matrix was used to obtain sensitivity, specificity and accuracy of the alternative sample treatment compared to the assays based on standard RNA extractions.
