Recognizing the potential danger of unforeseen pandemics, the Defense Advanced Research Projects Agency (DARPA) and National Institutes of Health (NIH) funded work in our laboratory to explore whether human organ-on-a-chip (Organ Chip) microfluidic culture technology might be helpful in confronting potential biothreat challenges. We previously showed that our Organ Chips can recapitulate human organ physiology, disease states, and therapeutic responses to clinically relevant drug pharmacokinetic (PK) exposures with high fidelity 4-8 . Here we show how human lung Airway Chips may be used to model human lung responses to viral infection in vitro, and in concert with higher throughput cell-based assays, to identify existing approved drugs that have the potential to be repurposed for treating or preventing spread of viral pandemics caused by influenza as well as the SARS-CoV-2 virus that is the cause of the current COVID-19 crisis.
