Snippet: The interferon induced protein with tetratricopeptide repeats (IFIT) protein family, present in all vertebrates, include some of the most highly expressed ISGs. IFIT1, IFIT2, IFIT3 and IFIT5 contain multiple IFN-stimulated response elements (ISREs) in their promoters (reviewed in 3) and are also induced directly by the IFN-regulatory factor 3 (4), downstream of initial PRR activation. Document: The interferon induced protein with tetratricopeptide repeats (IFIT) protein family, present in all vertebrates, include some of the most highly expressed ISGs. IFIT1, IFIT2, IFIT3 and IFIT5 contain multiple IFN-stimulated response elements (ISREs) in their promoters (reviewed in 3) and are also induced directly by the IFN-regulatory factor 3 (4), downstream of initial PRR activation. In contrast, the promoter of the poorly characterised human IFIT1B lacks ISREs. Different species have varying complements of IFITs, but most mammals possess IFIT1, IFIT1B, IFIT2, IFIT3 and IFIT5 (5) . Phylogenetic analysis revealed that rodents, including mice, have lost IFIT1 and instead Ifit1b1 is IFN responsive (6) . In mice Ifit1b1 has undergone duplication twice (Ifit1b2 and Ifit1b3) and Ifit3 once (Ifit3b) (6) . IFITs are composed of sequential tetratricopeptide repeat (TPR) motifs that form globular N-and C-terminal domains joined by a linker of variable flexibility (7) (8) (9) (10) .
