IFIT1 mutants were combined with IFIT3, reporter mRNA and RRL as described in Materials and Methods and Fluc dependent luminescence was measured ( Figure 5G ). At 40 nM IFIT1, 'YxxxL' motif mutants displayed similar translation inhibition. IFIT3 significantly enhanced translation inhibition of both wild type IFIT1 and the IFIT1-L464E mutant. In contrast, IFIT3 did not stimulate translation inhibition of either the IFIT1-Y460E mutant or the L464E/Y460E double mutant.
