Snippet: In the present study, we confirm that primary human NK cells treated with a combination of IL-33 and IL-12 ex vivo produce high levels of IFN- 16 and we extend this observation to near physiological picogram concentrations of these cytokines. Document: In the present study, we confirm that primary human NK cells treated with a combination of IL-33 and IL-12 ex vivo produce high levels of IFN- 16 and we extend this observation to near physiological picogram concentrations of these cytokines. Mechanistically, we implicate the p38 mitogen-activated protein kinase (MAPK) pathway in IL-33-mediated enhancement of IL-12-induced responses of human NK cells and uncover a role for ADAM17 in enhanced release of TNF under these stimulatory conditions. These data provide mechanistic insights into how IL-33-rich inflammatory milieus may directly enhance proinflammatory activities of NK cells.
