Additionally, replacing S27a 3'UTR with another unstructured 3' UTR of the same length significantly decreased protein expression (Supplementary Table 3 and Supplementary Fig. 1 ). Since the length of S27a 3' UTR (34nt) was considerably shorter than other 3' UTRs (AAT 100nt, TF 386nt, or HCV 190nt), S27a 3' UTR was used in the following studies.
