Title: Live-cell single RNA imaging reveals bursts of translational frameshifting Document date: 2018_11_24

Snippet: To further characterize the efficiency of frameshifting, we quantified the precise number of frameshifted versus non-frameshifted ribosomes per translation site. To do this, we re-imaged the MF tag at high laser powers so we could simultaneously visualize single mature XXLb1 or AlexX proteins in comparison to their nascent chains within translation sites (Fig. S4 ). Document: To further characterize the efficiency of frameshifting, we quantified the precise number of frameshifted versus non-frameshifted ribosomes per translation site. To do this, we re-imaged the MF tag at high laser powers so we could simultaneously visualize single mature XXLb1 or AlexX proteins in comparison to their nascent chains within translation sites (Fig. S4 ). After renormalizing the fluorescence, we found frameshifting sites with translation in both the 0 and -1 frames had 7.8 ± 0.7 ribosomes total, 1.9 ± 0.3 of which were frameshifted, while 0-frame only translation sites had 3.2 ± 0.1 ribosomes and -1-frame only translation sites had 3.4 ± 1.4 frameshifted ribosomes (Fig. 2B,D) .
