We also observed activation of PAR-2 by human lung tryptase and Der p 3 (Supplementary Fig S6) . PAR-2 activation also had a small but significant effect of acutely reducing squamous cell line metabolism and/or proliferation through lowering of cAMP, but did not activate apoptosis (Supplementary Fig S7) . PAR-2 activation increased secretion of TNFa and GM-CSF from RPMI 2650 cells and IL-6 and GM-CSF from NCI-H520 cells, likely involving dual coupling to Gq and Gi G protein isoforms (Supplementary Fig S8) .
