Title: Multiplex logic processing isothermal diagnostic assays for an evolving virus Document date: 2018_9_23

Snippet: Our results demonstrate that both the singleplex NS1 LAMP-OSD assay and the multiplex 4-Plex-LAMP-2GO assay could detect as few as 2 x 10 4 /ml of ZIKV virions in human saliva within 90 min (Figure 9 ). Since only a 1/200 fraction of this saliva sample was used in the nucleic acid tests, it is evident that our LAMP-based assays were able to generate positive signal from only 100 ZIKV virions. Saliva containing higher concentration of ZIKV virions.....
Document: Our results demonstrate that both the singleplex NS1 LAMP-OSD assay and the multiplex 4-Plex-LAMP-2GO assay could detect as few as 2 x 10 4 /ml of ZIKV virions in human saliva within 90 min (Figure 9 ). Since only a 1/200 fraction of this saliva sample was used in the nucleic acid tests, it is evident that our LAMP-based assays were able to generate positive signal from only 100 ZIKV virions. Saliva containing higher concentration of ZIKV virions yielded faster results with both assays. A typically encountered physiological viral load (2 x 10 5 virions / ml) generated bright fluorescence in the singleplex NS1 LAMP-OSD assay within only 30 min. The multiplex 4-Plex-LAMP-2GO assay required 60 min to generate discernible signal with the same sample. Neither assay generated non-specific fluorescence when challenged with saliva containing as many as 10 7 /ml of non-specific virions.
