The cells are transfected with mRNA by adding mRNA nanocarriers using a perfusion system and incubating the complexes for one hour before the fluid is exchanged with cell growth medium (the two perfusion steps and the incubation is indicated by the arrows and the gray bar). Subsequently, we record the fluorescent protein expression time courses. Representative single-cell time courses illustrate the variation for protein expression levels (B) and eGFP expression onset times (C).
