The following modifications were made to the original protocol: 400 µl of lysis buffer, 400 µl of binding buffer and 20 µl of proteinase K and carrier RNA were used per sample. Samples were eluted in 60 µl. The NucleoSpin Soil kit (Macherey-Nagel) was used to extract DNA/RNA from sediment. 500 mg of soil was extracted according to the manufactures protocol using an elution volume of 100 µl.
