Document: Although the antibacterial and tumoricidal roles of γδ T cells have been well studied, their roles during influenza virus infection remain poorly understood. In this study, we demonstrate for the first time that γδ T cells could be directly activated by HPAI H5N1 viruses and that γδ T cells play an important role in the defense against HPAI H5N1 infection. The trimers but not monomers of HPAI H5N1 viral HA proteins could also directly activate γδ T cells, and this activation depends on sialic acid receptors on the surface of γδ T cells and on the glycosylation of HA proteins. Moreover, HA-induced γδ T cell activation could be inhibited by the phosphatase calcineurin inhibitor CsA. These findings provide a further understanding the mechanism underlying γδ T cell-mediated innate and adoptive immune responses against HPAI H5N1 viral infection.
