Briefly, NSG mice (4-to 6-week old) were implanted with 1 mm 3 sections of fetal thymus and liver (Advanced Biomedical Resources, Alameda, CA) under the kidney capsule. Two weeks post-implantation the mice were treated with busulfan (Sigma) (20 mg/kg IP) and were injected retro-orbitally with 5x10 5 CD34 + stem cells, isolated from the donor fetal liver using MACS, (Miltenyi Biotec Inc. Auburn, CA). Eight weeks following the stem cell xenograft the peripheral blood from the BLT mice was screened for the presence of human cells. BLT mice were screened and selected using the same protocol described above for the CD34 + cord blood mice.
