Document: Lower transformation rates compared to E. coli must be considered for antibody library generation rather than for antibody production. Moreover, the frequency of homologous transformation in yeast is higher compared to higher eukaryotes facilitating the process of making stable expression clones. Specific issues of heterologous protein expression in yeast can be circumvented by optimizing gene sequences, for example by avoiding AT-rich stretches which can cause premature transcriptional termination. The productivity of antibody fragments in yeasts was increased by DNA shuffling (106).
