The internal standard has a different base composition and thus can be differentiated. As this synthetic standard is added to each PCR well at a known copy number, the comparison between standard and microbial DNA permits quantification. In the absence of a PCR product, the internal standard serves as PCR positive control to exclude PCR inhibition.
