    Document: In this study, we have investigated the organization of the MW and further evaluated the role of the nuclear transport machinery in the formation of a selective barrier between the viral replication/assembly complexes and the cytosol. We present data supporting a role for virusinduced cytoplasmic compartments in concealing viral PAMPs from RLR recognition in the cytoplasm of cells infected with positive-strand RNA viruses. We also provide evidence for the formation of distinct replication and assembly compartments within the MW, and we describe a useful technique for isolating these compartments. Furthermore, we present data indicating the nuclear transport machinery contributes to the selective movement of molecules into the MW and the exclusion of cytoplasmic RLRs.
