Herein, we demonstrate that poly(I:C) stimulation of mixed glial cultures results in a reduction of oligodendrocyte precursor (preOL) viability in a TNFα-and TNFR1-dependent mechanism. Interestingly, the effects of poly(I:C) stimulation were very similar to those induced by LPS stimulation, and suggest that without proper regulation of glial inflammatory responses, aberrant production of TNFα within the CNS can be detrimental to developing oligodendrocytes.
