GFP-expressing C4-2 cell line was used as a model because of the advantages of fluorescence imaging techniques such as microscopy and quantitative flow cytometry. Figure 3a -f shows confocal images of the C4-2 cells without treatment, treated with GFP-siRNA alone, chimera alone, a random sequenced siRNA with the protein tag (His 18 ), chimera with protein tag (His 6 ), and chimera with protein tag (His 18 ). Qualitatively, only the experimental treatment, chimera with protein tag (His 18 ), clearly shows GFP silencing, whereas none of the five control treatments leads to significant suppression of GFP expression.
