Blood was incubated with six concentrations of aspirin and aggregation was measured using a multiple electrode impedance aggregometer (Multiplate instrument). In vivo testing was performed at least 1 week after in vitro testing. The same dogs received a single dose of 1 mg/kg aspirin orally and aggregation was assessed at baseline prior to administration, then 20 minutes, 40 minutes, and 180 minutes after administration. Additionally, plasma concentrations of ASA and salicylic acid (SA) were measured at each time point using liquid chromatography-mass spectrometry (LC-MS). SA is the major ASA metabolite and may be a better indicator of ASA exposure (bioavailability) than ASA.
