Employing the optimized SpyTag/SpyCatcher system, we have assembled vaccines composed of a single-chain antibody against DEC205 and model antigens (including the model T-cell epitope chicken ovalbumin 257-264 (OVA 8 ) and tick-borne encephalitis virus envelope protein domain 3 (TBEV ED3)) 22, 23 . This new synthetic vaccine was shown to be fully functional and to generate efficient cytotoxic T-cell and antibody responses. Thus, this protein-based synthetic vaccine strategy may be a significant improvement over the conventional gene-based synthetic vaccine strategy and may serve as a useful platform for faster and easier vaccine development.
