The reversible inhibitor of Golgi tx-mannosidase I, dMM (4), was included in the labeling and chase media, so that man6P receptors within and beyond the TGN would bear high-mannose oligosaccharides. Next, dMM was washed away, and cells were incubated for various lengths of time in the presence or absence of BFA. We then monitored the potential galactosylation of man6P receptors that would occur if they returned to the ER.
